Subcellular localization of P-glycoprotein in the biliary epithelium of human bile ducts: effect of biliary diseases on the expression. We examined the distribution of P-glycoprotein (P-gp) in the biliary epithelium of human bile ducts and the effect of biliary diseases on the expression of P-gp. Immunohistochemical examination demonstrated that the biliary epithelium of both normal ducts and cholangiocarcinomas strongly expressed P-gp. The intensity of immunostaining was relatively lower in the intrahepatic bile ducts than in the extrahepatic bile ducts. The cholangiocellular carcinomas showed a weaker expression of P-gp than the adenocarcinomas. In situ hybridization revealed the distribution of mRNA for P-gp in both the plasma membrane and the cytoplasm of the biliary epithelium, whereas the intraluminal contents of the bile ducts did not show specific signal. Western blotting and reverse transcription-polymerase chain reaction analysis revealed that the expression of P-gp was significantly reduced in the cholangiocarcinoma cells as compared with that in the intrahepatic bile ducts. These findings suggest that the expression of P-gp in the biliary epithelium is regulated by the diseases of the bile ducts and is associated with the intracellular transport of drugs and biliary diseases.(Reuters) - Consumer products retailer Dollar General Corp DG.N and its subsidiaries agreed to be acquired by wholesale retailer Alliance Co Ltd 2315.T for a price of $6.2 billion in cash, people familiar with the matter said on Thursday.
